Active Vitamin D supports bone cell precursor recycling and development by increasing BECN1 gene activity

Updated

Abstract

1α,25-(OH)2-D3 promotes the expression of BECN1 mRNA and Beclin1 protein in osteoclast precursors.

  • 1α,25-(OH)2-D3 enhances autophagy of osteoclast precursors, which may limit its effectiveness against osteoporosis.
  • Increased p65 phosphorylation and nuclear translocation are observed following treatment with 1α,25-(OH)2-D3.
  • Silencing BECN1 and p65 disrupts the effects of 1α,25-(OH)2-D3 on osteoclast differentiation and autophagy.
  • Silencing p65 also prevents the increase in BECN1 mRNA and Beclin1 protein levels induced by 1α,25-(OH)2-D3.
  • Targeted inhibition of BECN1 or p65 may enhance the anti-osteoporotic effects of 1α,25-(OH)2-D3.
