17α-Estradiol Provides Limited Protection Against APOE4-Related Traits in Middle-Aged Female Mice

Treatment with 17α-estradiol for 6 months shows modest improvements in systemic aging phenotypes in female mice, particularly those with the APOE4 genotype.

  • APOE4 genotype in female mice exhibits more severe aging-related traits, such as increased body fat and impaired glucose tolerance, compared to APOE3.
  • 17α-estradiol treatment results in decreased body fat and increased lean mass, glucose tolerance, and energy expenditure, primarily benefiting APOE4 females.
  • Behavioral performance varies with APOE genotype; APOE4 females perform worse in memory tasks and show higher levels of the Alzheimer's-related peptide soluble β-amyloid.
