[18F]DPA-714 levels in blood vary with other medicines, age, sex, body weight, and genetic differences in a key protein

Abstract

In a cohort of 201 subjects, [F]DPA-714 metabolism showed significant interindividual variability, with some subjects experiencing changes in concentration of up to 88%.

  • [F]DPA-714 levels were similar between patients and healthy controls, with values of 59.7 ± 12.3% and 60.2 ± 12.9%, respectively.
  • Subjects receiving co-medications that inhibit or induce CYP3A4 exhibited drastic changes in [F]DPA-714 levels, with increases of up to 88% and decreases down to 23%.
  • A significant bias of about 30% in the cortex-to-plasma ratio was observed when not considering individual metabolism rates.
  • Metabolism of [F]DPA-714 decreased with age and body mass index, and was faster in females than in males.
  • High uptake of [F]DPA-714 was noted in organs rich in the translocator protein, with marked accumulation in plasma for low-affinity binders.
