3-TYP may protect against heart failure with normal pumping by blocking Sirtuin 3

Abstract

Sirtuin 3 (Sirt3) protein expression significantly increased in spontaneously hypertensive rats (SHRs) with heart failure with preserved ejection fraction (HFpEF).

  • High-fat feeding and streptozotocin injection in SHRs resulted in impaired diastolic function and increased cardiac hypertrophy and fibrosis.
  • Inhibition of Sirt3 with 3-TYP led to significant improvements in cardiac function and a reversal of hypertrophic changes.
  • Increased Sirt3 expression is associated with elevated endoplasmic reticulum stress and mitochondrial autophagy under metabolic stress.
  • Silencing Sirt3 markedly reduced excessive endoplasmic reticulum stress and levels of mitophagy induced by metabolic stress.
  • 3-TYP may serve as a therapeutic candidate for managing HFpEF in patients with metabolic syndrome.
