Activation of serotonin 5-HT2A receptors causes long-lasting presynaptic inhibition in the reward system depending on cannabinoid CB1 receptors

Updated

Abstract

Activation of 5-HT2A receptors induced a robust inhibitory long-term depression (iLTD) of evoked GABAergic synaptic transmission.

  • 5-HT2A receptor activation may modulate GABAergic synaptic transmission onto medium spiny neurons in the nucleus accumbens.
  • iLTD was associated with reduced spontaneous and miniature inhibitory postsynaptic current (IPSC) frequency.
  • An increased paired-pulse ratio and changes in release probability suggest a presynaptic reduction in GABA release.
  • Endocannabinoid signaling is critical for iLTD, as indicated by the effects of the CB1 receptor agonist and antagonist.
  • iLTD induction requires postsynaptic G-protein signaling, phospholipase Cβ activity, and synthesis of the endocannabinoid 2-arachidonoylglycerol.
