How 5-methoxyindole products of L-tryptophan may control COX-2 levels, inflammation, and tumor growth

Abstract

5-methoxytryptophan (5-MTP) inhibits COX-2 transcriptional activation, which is associated with inflammation and tumor growth.

  • 5-MTP is produced by fibroblasts and can restore control of COX-2 overexpression in cancer cells.
  • Cancer cells exhibit a deficiency in producing 5-MTP, contributing to persistent COX-2 overexpression.
  • 5-MTP blocks cancer cell migration and invasion in laboratory studies.
  • In a xenograft model, 5-MTP inhibits tumor growth and cancer metastasis.
  • Melatonin also suppresses COX-2 and exhibits anti-cancer properties, but requires higher concentrations.
  • 5-hydroxyindole metabolites of L-tryptophan do not control COX-2 expression.
