A Small Molecule That Detects Damaged Mitochondria and Selectively Boosts Their Removal by Autophagy

Updated

Abstract

MG-1 markedly enhanced autophagic flux and mitochondrial clearance under conditions of mitochondrial stress.

  • Bifunctional small molecules can activate damage-responsive mitophagy by linking mitochondrial recognition with autophagic machinery.
  • MG-1 was identified as a lead compound with minimal effects on normal autophagy.
  • Biochemical analyses confirmed that MG-1 promotes the breakdown of damaged mitochondria through lysosomes without directly uncoupling mitochondria.
  • MG-1 demonstrated good tolerance in experimental systems and improved movement in a zebrafish model of mitochondrial dysfunction.
