Single Injection of AAV2-FGF18 Gene Therapy May Reduce Cartilage Loss and Bone Damage in a Mechanical Osteoarthritis Model

Updated

Abstract

Total defect width in joints treated with AAV2-FGF18 was reduced by up to 76% compared to controls.

  • AAV2-FGF18 treatment led to significant reductions in cartilage defect size and increased cartilage thickness.
  • Vehicle-treated joints displayed severe degeneration, including cartilage erosion and bone void formation.
  • AAV2-FGF18-treated joints appeared more anatomically normal with only minor regional damage.
  • Injections of rh-FGF18 resulted in joint swelling and changes in cartilage protein expression.
  • Gene therapy treatment showed durability and localized effects, upregulating chondroanabolic genes while suppressing markers associated with osteoarthritis.
