Improving plasmid DNA recovery using membrane chromatography with added chemicals

Updated

Abstract

Plasmid recovery increased from approximately 50% to as high as 93% using optimized membrane chromatography.

  • Optimized parameters included pH, conductivity, loading conditions, and buffer additives.
  • Arginine and urea significantly enhanced plasmid recovery and resolution between pDNA and RNA.
  • Additives reduced the conductivity needed for plasmid elution, improving recovery and selectivity.
  • Similar benefits were observed with guanidine and histidine as additional enhancers.
  • The approach enables rapid processing of clarified lysate without extra chromatography steps.
