Fat tissue stem cell fluid helps skin cells at different aging stages resist UVB damage

Abstract

ADSC-conditioned medium (ADSC-CM) can improve cellular proliferation and reduce senescence in irradiated human dermal fibroblasts (HDFs).

  • ADSC-CM pretreatment led to increased expressions of collagen I, collagen III, elastin, and TIMP-1 in irradiated HDFs.
  • A decrease in the expressions of MMP-1 and MMP-9 was observed in both irradiated and nonirradiated HDFs after ADSC-CM treatment.
  • The concentration of platelet-derived growth factor AA (PDGF-AA) in ADSC-CM decreased over time but maintained bioactivity for up to 11 weeks at low temperature.
  • Significant increases in pAkt protein expression were noted after ADSC-CM pretreatment, indicating activation of the PI3K/Akt signaling pathway.
  • The effects of ADSC-CM pretreatment were consistent across three generations of HDFs, although variations in results were observed.
