Abstract
Over 96% of contaminants are removed from isolated extracellular vesicles (EVs) using a novel engineering approach.
- Structure-guided engineering of tetraspanins CD63 and CD9 allows for the efficient isolation of intact EVs.
- The internal insertion of a twin-Strep tag facilitates antibody-free affinity isolation of EVs.
- Proteins of interest can be loaded into EVs with optogenetic control over their release.
- The engineered EVs maintain high purity, enabling the detection of potentially novel EV-associated proteins.
- This method effectively removes hepatitis C virus and adeno-associated virus, indicating high specificity.
- Coating EVs with vesicular stomatitis virus glycoprotein enhances their ability to escape endosomes.
