Age-related DNA changes in mouse blood and tail: possibility of creating tissue-specific biological clocks

Abstract

A total of 78,111 age-associated regions of DNA methylation were identified in blood and 38,348 in tail tissues from mice aged 7-103 weeks.

  • Blood samples exhibited predominantly hypomethylating changes with a more abrupt age transition compared to tail samples.
  • Tail tissues demonstrated a balance of hypermethylation and hypomethylation with smoother age-related changes.
  • Segment-level analysis revealed 31,322 differentially methylated regions in blood and 21,883 in tail tissues.
  • Out of the identified regions, 6229 were shared between blood and tail, showing specific patterns of hyper- and hypomethylation.
  • Elastic-net models for predicting age achieved high accuracy, with blood samples showing a mean absolute error of 2.30 weeks.
  • The blood clock also demonstrated some generalizability to an independent dataset, though with a higher mean absolute error of 13.48 weeks.
