SIRT2's protective role in diet-induced fatty liver disease in diabetic rats shown by AGK2 and nicotinamide riboside through regulating inflammation, energy control, and cell defense pathways

Updated

Abstract

Nicotinamide riboside (NR) treatment improved fasting glucose and reduced steatosis in a diabetic rat model of non-alcoholic steatohepatitis (NASH).

  • SIRT2 activation through NR led to improvements in insulin resistance and oral glucose tolerance.
  • Histological analysis showed decreased inflammation, oxidative stress, and fibrosis in treated subjects.
  • NR treatment enhanced SIRT2-mediated reduction of inflammatory markers and restored AMPK signaling.
  • Pharmacological inhibition of SIRT2 with AGK2 diminished the beneficial effects of NR on metabolic and hepatic outcomes.
  • Data suggest that SIRT2 activation may serve as a protective mechanism against metabolic liver disease.
