Using AI to design a COVID-19 spike protein that may protect against many related coronaviruses

Updated

Abstract

Engineered Spike immunogens elicited robust antibody responses in mice and demonstrated broad neutralizing activity against diverse SARS-CoV-2 variants and zoonotic sarbecoviruses.

  • A structure-guided, AI-enabled strategy for antigen design was utilized to enhance cross-reactive B-cell epitope recognition across betacoronaviruses.
  • Conserved epitope hotspots within the Spike receptor-binding domain were identified through comparative sequence analysis and conformational epitope prediction.
  • Targeted changes were made to a SARS-CoV-2 BA.2 backbone to improve antibody responses.
  • Selected designs exhibited neutralization breadth comparable to an Omicron-adapted clinical benchmark for shared SARS-CoV-2 antigens.
  • The engineered vaccines extended neutralizing activity to divergent zoonotic sarbecoviruses.
