Abstract

Ajugol administration significantly reduced inflammatory cytokines and NLRP3 expression in MSU-induced gouty arthritis mice.

  • Gouty arthritis is triggered by the deposition of monosodium urate crystals and is linked to oxidative stress and mitochondrial dysfunction.
  • Network pharmacology identified the PI3K/AKT/mTOR pathway as a critical regulatory pathway in gouty arthritis.
  • Ajugol was found to restore cell viability and preserve extracellular matrix integrity in LPS/MSU-stimulated human chondrocytes.
  • Ajugol inhibited the expression of key components involved in pyroptosis, including NLRP3 and caspase-1, while reducing secretion of various inflammatory cytokines.
  • The activation of mitophagy by ajugol was confirmed to play an essential role in its protective effects against gouty arthritis.
  • In vivo studies demonstrated that ajugol alleviated joint swelling and preserved cartilage integrity through the suppression of the PI3K/AKT/mTOR pathway.
