Autophagy

A specific protein modification helps build a gene-regulating complex that controls cell cleanup and protein balance in non-dividing eye support cells

Abstract

AKT1 drives autophagy through a non-canonical mechanism in retinal pigment epithelial cells.

  • AKT2 hyperactivation destabilizes MTORC2 and inhibits IRS-1, resulting in autophagic arrest.
  • Compensatory activation of AKT1 leads to the phosphorylation of TERT, promoting its nuclear translocation.
  • In the nucleus, TERT forms a complex with FOXO3 and MYC that activates the EIF2AK3 promoter.
  • This activation shifts the unfolded protein response from pro-apoptotic to cytoprotective, promoting autophagy machinery and selective ERphagy.
  • AKT1-mediated phosphorylation of TERT is necessary for nuclear function of FOXO in post-mitotic cells.
