Alcohol disrupts a key regulator in liver cells through FoxO1, promoting cell death by increasing TXNIP levels

Abstract

A significant decrease of miR-148a was observed in the liver of patients with alcoholic hepatitis.

  • Alcohol exposure led to reduced levels of miR-148a in both human and mouse liver samples.
  • Decreased miR-148a levels were associated with inhibited expression of a key transcription factor, FoxO1.
  • Overexpression of thioredoxin-interacting protein (TXNIP) was linked to alcohol-induced hepatocyte damage.
  • Activation of the NLRP3 inflammasome and pyroptosis in hepatocytes was triggered by increased TXNIP levels.
  • Restoring miR-148a levels or inhibiting TXNIP reversed the detrimental effects of alcohol on liver cells.
