Energy and recycling signals control iron-related cell death caused by BPA in hippocampal nerve cells

Updated

Abstract

BPA exposure caused oxidative damage and mitochondrial ultrastructural abnormalities in HT-22 hippocampal neuronal cells.

  • BPA exposure led to increased levels of malondialdehyde (MDA), PTGS2 protein, and reactive oxygen species (ROS).
  • Concomitantly, levels of glutathione (GSH), glutathione peroxidase 4 (GPX4), and SLC7A11 decreased.
  • Ferroptosis inhibitors, ferrostatin-1 and deferoxamine, reversed BPA-induced changes.
  • Inhibition of autophagy with chloroquine reversed alterations in GPX4, SLC7A11, and PTGS2 caused by BPA.
  • Decreased expression of nuclear receptor coactivator 4 (NCOA4) and ferritin heavy chain 1 (FTH1) was observed with BPA exposure.
  • BPA activated the AMPK/ULK1 signaling pathway while inhibiting mTOR, suggesting a role in promoting ferroptosis.
