Antibody responses after COVID-19 vaccination and breakthrough infections show lasting memory of the original virus strain in both first-time and recovered individuals

Abstract

Titers of neutralizing antibodies following primary vaccination with adenovirus vectored vaccine were significantly lower than those following mRNA vaccination.

  • An mRNA vaccine booster dose resulted in equivalent binding and neutralizing antibody titers to the ancestral virus for all individuals, regardless of the primary vaccine type.
  • Vaccinated infection-naïve individuals required a third dose of mRNA vaccine to produce neutralizing antibodies against Omicron subvariants BA1, BA2, and BA5.
  • Neutralizing antibody titers against Omicron variants were lower than those against the ancestral strain.
  • Breakthrough infection with Omicron strains led to a greater increase in neutralizing antibody titers against the ancestral strain compared to Omicron variants.
  • Evidence of antigenic imprinting was observed, indicating a stronger immunologic response to the ancestral vaccine antigen than to the infecting Omicron variant.
