Abstract
Liposomal anti-PD1 formulation (LAP) achieved complete tumor rejection in 67% of mice, compared to 17% with conventional antibody treatment.
- LAP demonstrated efficient binding to targets with a mean particle size of 89 nm.
- Intravenous administration of LAP resulted in a rapid but temporary increase in circulating pro-inflammatory cytokines.
- Tumor growth inhibition with conventional anti-PD1 therapy was modest and not statistically significant.
- LAP significantly reduced tumor growth and improved survival in the B16-F10 melanoma model.
- LAP altered the tumor environment by reducing pro-tumoral M2-tumor-associated macrophages and increasing the ratio of cytotoxic to regulatory T-cells.
- Depletion of myeloid (CD11b+) cells reduced the antitumor effectiveness of LAP but did not significantly change pharmacokinetics.
