How ANXA3-related cell stress may encourage inflammatory immune cell changes in lung artery high blood pressure using bioinformatics and machine learning

Abstract

Fifty differentially expressed endoplasmic reticulum stress-related genes were identified, with four key candidates validated in pulmonary arterial hypertension (PAH) models.

  • Four key genes associated with endoplasmic reticulum stress were identified: BCL2L1, TNFRSF17, DNAJB1, and ANXA3.
  • ANXA3 was found to be overexpressed in macrophages associated with PAH and co-localized with inflammatory macrophages in lung tissues.
  • Silencing ANXA3 improved right ventricular function and reduced pulmonary vascular remodeling in animal models.
  • In vitro studies showed that ANXA3 knockdown suppressed the inflammatory response in macrophages by decreasing cytokine secretion and inhibiting markers of endoplasmic reticulum stress.
  • ANXA3 knockdown also reduced the proliferation and migration of pulmonary artery smooth muscle cells triggered by inflammatory stimuli.
