Abstract

A higher ApoE4 allele count is linked to lower plasma ptau217 thresholds for early pathological tau-PET increases.

  • ApoE4 is associated with faster tau-PET spread from local brain regions in a manner dependent on the number of ApoE4 alleles.
  • There is no evidence that ApoE4 facilitates amyloid-PET-associated increases in plasma ptau217.
  • Plasma ptau217 elevations correlate with increased tau aggregation across brain areas, independent of amyloid levels.
  • The study analyzed two large cohorts (ADNI: n=201, A4-LEARN: n=200) to assess the relationship between ApoE4 and tau pathology.
  • Findings indicate that ApoE4 may enhance the spread of tau driven by phosphorylated tau, leading to an earlier transition from amyloidosis to tauopathy.
