Activating cell cleanup processes reduces daily clock rhythms in U2OS cells through the ATG5 pathway

Updated

Abstract

Autophagy activation significantly decreases the expression of circadian clock genes in U2OS cells.

  • The mRNA levels of autophagy-related genes MAP1LC3B and ATG5 are significantly increased following autophagy activation.
  • Circadian clock genes, including BMAL1, PER2, REV-ERBα, and DBP, show significantly decreased expression after autophagy activation.
  • Both untreated and treated U2OS cells exhibit rhythmic expression patterns of circadian clock and autophagy-related genes.
  • Inhibition of autophagy with 3-methyladenine partially reverses the decrease in circadian clock gene expression.
  • Knockout of ATG5 reduces the negative impact of autophagy activation on circadian clock oscillators.
