Activating cell cleaning in inflamed tissues may improve therapy with stem cell vesicles in lupus

Updated

Abstract

Rapa-SLE-EV administration significantly ameliorated autoimmune response and renal damage in MRL/lpr mice.

  • Autophagy was triggered in MSCs by interaction with the SLE inflammatory microenvironment and enhanced by rapamycin treatment.
  • Rapa-SLE-EVs outperformed other MSC-derived EVs in reducing autoantibody levels, splenomegaly, and lymphadenopathy.
  • This treatment suppressed plasma inflammatory cytokines and preserved renal function while mitigating pathological damage.
  • Rapa-SLE-EV exhibited strong inhibitory effects on SLE-B cell function, attributed to high levels of the anti-inflammatory protein IDO1.
  • IDO1 was confirmed to enter SLE-B cells via the extracellular vesicles.
