Lipid nanoparticles with bile acids improve mRNA delivery outside the liver

Abstract

Bile acid-containing lipid nanoparticles (BA-LNPs) can reduce delivery to liver cells and enhance delivery to other cell types.

  • Incorporating bile acids into lipid nanoparticles allowed for improved targeting of mRNA to T cells, B cells, and epithelial cells.
  • A specific formulation, CA-100, demonstrated significant tropism for the spleen when administered intraperitoneally or intravenously.
  • Replacing cholesterol with bile acids in the formulation led to a shift in mRNA delivery from the liver to the spleen.
  • BA-LNPs may offer a novel strategy for extrahepatic delivery of mRNA therapeutics in various applications.
