Abstract

MnNE-mOVA significantly suppressed tumor growth and prolonged survival compared with LNP-based formulations in E.G7-OVA tumor models.

  • A PEG-free nanoemulsion platform was developed to deliver mRNA and reduce liver accumulation.
  • Manganese-modified nanoemulsion (MnNE) effectively activated the STING pathway.
  • MnNE demonstrated superior transfection efficiency in antigen-presenting cells compared to the Moderna LNP formulation.
  • Strong antigen-specific humoral and cellular immune responses were elicited using ovalbumin as a model antigen.
  • MnNE-mOVA was associated with the most effective reversal of the immunosuppressive tumor microenvironment among treatment groups.
