Bivalent S-EABR mRNA-LNP boosters elicited the highest neutralizing antibody titers against Omicron subvariants in mice with pre-existing immunity.

  • The EABR mRNA vaccine platform enhances immune responses by presenting engineered immunogens on cell surfaces.
  • Monovalent and bivalent S-EABR mRNA-LNP boosters improved neutralizing responses against multiple Omicron subvariants compared to conventional boosters.
  • Bivalent S-EABR mRNA-LNP consistently resulted in higher antibody titers than monovalent formulations.
  • Epitope mapping indicated that bivalent S-EABR mRNA-LNP boosted a diverse range of anti-RBD responses, targeting multiple epitope classes.
  • Cryo-EM analysis suggested that bivalent immunizations promote the formation of spike protein structures that may enhance antibody diversity.
