PD-L2 is upregulated in senescent human cells and during aging.

  • Senescent cells accumulate with age and may contribute to metabolic dysfunction and reduced fitness.
  • Senolytics, which target senescent cells, can remove those that express high levels of PD-L2.
  • Old mice lacking PD-L2 show fewer senescent cells and improved insulin sensitivity and grip strength.
  • Treatment with anti-PD-L2 therapy can restore insulin sensitivity in aged mice.
  • PD-L2 functions as an immune checkpoint, helping senescent cells avoid immune clearance.
