PD-L2 is upregulated in senescent human cells and during aging.
- Senescent cells accumulate with age and may contribute to metabolic dysfunction and reduced fitness.
- Senolytics, which target senescent cells, can remove those that express high levels of PD-L2.
- Old mice lacking PD-L2 show fewer senescent cells and improved insulin sensitivity and grip strength.
- Treatment with anti-PD-L2 therapy can restore insulin sensitivity in aged mice.
- PD-L2 functions as an immune checkpoint, helping senescent cells avoid immune clearance.
