Abstract

In a sample of 48,524 participants, plasma neurofilament light chain (NfL) was associated with an increased risk of incident amyotrophic lateral sclerosis (ALS) with a hazard ratio of 1.69.

  • NfL was linked to higher risks of incident atypical parkinsonian disorders (APD), Alzheimer's disease (AD), and Parkinson's disease (PD) with hazard ratios of 1.51, 1.31, and 1.14, respectively.
  • GFAP showed an independent association with incident AD only, with a hazard ratio of 1.72.
  • The findings highlight distinct pre-diagnostic profiles for NfL and GFAP in relation to neurodegenerative diseases.
