Abstract

In a cohort of over 500,000 UK Biobank participants, platelet factor 4 (PF4) was identified as a key regulator associated with ALS risk.

  • Platelet indices were strongly linked to the risk of developing ALS.
  • Serum levels of PF4 were significantly lower in patients diagnosed with ALS.
  • Administering recombinant PF4 in mouse models of ALS led to extended survival and improved motor function.
  • PF4 treatment reduced neuroinflammation and neuromuscular junction denervation in specific ALS models.
  • The therapeutic effects of PF4 appear to be selective for SOD1-driven ALS models, with no notable impact on other ALS types.
  • PF4 activates the TBK1-OPTN signaling pathway, restoring autophagic processes without relying on PINK1 or Parkin.
