Using Network Models to Find Brain Resilience in a Mouse Model of Early Alzheimer's Without Symptoms

Updated

Abstract

A novel preclinical model of Alzheimer’s disease (AD) reveals that male CgA-KO/PS19 mice develop AD-like profiles while retaining intact learning and memory.

  • Approximately 20-30% of older adults exhibit AD hallmark lesions without cognitive impairment, termed asymptomatic AD (AsymAD).
  • Boolean network modeling identified a consistent gene signature that stratifies AD disease states across various datasets.
  • In the pre-frontal cortex of male CgA-KO/PS19 mice, AD-like molecular changes were observed without cognitive decline.
  • Female CgA-KO/PS19 mice showed even greater resilience, marked by reduced Tau phosphorylation and preserved synaptic structure.
  • CgA is identified as a key factor that may connect neuroendocrine signaling, Tau pathology, and cognitive preservation.
