Reducing bone loss in rheumatoid arthritis by blocking BRD4 to lower bone-breakdown cell activation caused by RANKL

Abstract

BRD4 expression significantly increased in the synovial tissues of RA patients and the ankle joints of CIA mice.

  • The BRD4 inhibitor JQ1 effectively reduced inflammation and arthritis severity (p < 0.05) in a collagen-induced arthritis mouse model.
  • Treatment with JQ1 improved bone mass and structural integrity in CIA mice while downregulating osteoclast-related gene expression.
  • JQ1 also inhibited the RANKL/RANK signaling pathway, indicating a reduction in bone erosion.
  • In vitro assays showed that JQ1 markedly inhibited osteoclast differentiation and function.
  • These findings suggest that BRD4 may play a pivotal role in bone destruction associated with rheumatoid arthritis.
