Abstract

A total of 42 senescence-related differentially expressed genes (DEGs) were identified in sarcopenia.

  • Significant involvement of kinase activity regulation and receptor tyrosine kinase-related signaling was observed in sarcopenia.
  • GSEA showed positive enrichment of extracellular matrix-related pathways and negative enrichment of the citric acid cycle in sarcopenia.
  • PCK1 and EGFR were significantly upregulated, while MAPKAPK3 was significantly downregulated in sarcopenia.
  • The combined three-gene model achieved an improved diagnostic performance with an AUC of 0.801.
  • PCK1 and EGFR were positively associated with fibroblast-related features, while MAPKAPK3 correlated with Th1-cell-related immune characteristics.
  • Experimental validation in a rat model supported the findings, showing significant changes in mRNA and protein levels of the identified hub genes.
