How Carbonic Anhydrase 9 Controls pH to Help Pancreatic Cancer Cells with KRAS Activation Survive Low Oxygen

Abstract

CA9 was expressed by 66% of pancreatic ductal adenocarcinoma (PDAC) samples analyzed.

  • PDAC cells under hypoxic conditions showed increased levels of HIF1A and HIF2A, leading to upregulation of CA9 and activated glycolysis.
  • Knocking down KRAS or using trametinib reduced stabilization of HIF1A and HIF2A, along with CA9 expression and glycolysis in PDAC cells.
  • High CA9 expression in PDAC samples correlated with significantly reduced survival times of patients.
  • Inhibition of CA9 in PDAC cells under hypoxic conditions decreased intracellular pH and enhanced sensitivity to gemcitabine.
  • PDAC cells with CA9 knockdown formed smaller tumors in mice, while combination treatment with SLC-0111 and gemcitabine resulted in increased intratumor acidosis and cell death.
