Repeated LSD and MDMA exposure may pose valvular heart risk through sustained 5-hydroxytryptamine 2B receptor signaling.

Evidence

This systematic review searched Google Scholar, OVID, and PubMed through June 1, 2025 and included 17 mechanistic, preclinical, structural, and human studies on psychedelic or MDMA links to valvulopathy pathways.

Caveat

No psilocybin, dimethyltryptamine, or mescaline studies and no clinical LSD-induced valvulopathy cases were found, leaving the strongest human signal limited to chronic MDMA use.
