Differences in levels of two protein enzymes in joint tissue from rheumatoid arthritis and osteoarthritis

Abstract

Cathepsin K protein expression in RA-derived synovial specimens was 2-5 times higher than in OA samples (P < 0.001).

  • Cathepsin K and S proteins were identified in the synovium of patients with rheumatoid arthritis (RA) and osteoarthritis (OA).
  • Cathepsin K was found in synovial fibroblasts and multinucleated giant cells, particularly at sites of cartilage erosion in RA.
  • Cathepsin S expression was limited to macrophage-like synoviocytes and blood vessel endothelial cells, with no significant differences between RA and OA.
  • RA and OA-derived synovial fibroblast cultures expressed similar levels of cathepsin K, with both forms of the enzyme secreted into the culture medium.
  • IL-1 beta and TNF alpha increased cathepsin K expression in fibroblast cultures by 7-8 times at the transcript level and 2 times at the protein level.
