Effects of positively charged porphyrins that interact with DNA structures on human cancer cells

Abstract

50% inhibition of telomerase activity was achieved at porphyrin concentrations of ≤ 50 microM.

  • Cationic porphyrins interact with and stabilize telomeric G-quadruplex DNA.
  • Cytotoxicity against normal and tumor cells was generally observed at concentrations > 50 microM.
  • Porphyrins were effectively absorbed into the nuclei of tumor cells in culture.
  • Inhibition of telomerase activity in MCF7 cells was dependent on time and concentration of TMPyP4 over a 15-day period.
  • Cell growth arrest occurred in the G2-M phase alongside telomerase inhibition.
