Two enzymes control fat droplet growth in cells using different methods

Updated

Abstract

Knockdown of CDS1 or CDS2 resulted in the formation of giant lipid droplets in cultured mammalian cells.

  • CDS1 and CDS2 are involved in lipid droplet biogenesis and growth through distinct mechanisms.
  • Down-regulation of a key regulator of lipid droplet growth restored lipid droplet size in specific deficient cells.
  • Reduction of two enzymes associated with triacylglycerol synthesis improved the lipid droplet phenotype in certain deficient cells.
  • Deficiency in specific enzymes promoted lipid droplet association of key synthesis proteins and impeded initial lipid droplet maturation.
  • CDS2 has a stronger impact than CDS1 on regulating phosphatidic acid levels on the surface of lipid droplets.
