Cell penetrating thyclotides (CPTs) significantly enhance the cellular uptake and endosomal escape of bioactive peptides.

- Peptides, despite their medicinal potential, often struggle to cross cell membranes effectively.
- CPTs combine chiral tetrahydrofurans and polar sidechains, allowing for optimization in peptide delivery.
- Two anticancer peptides with low bioavailabilities showed markedly improved cellular uptake when conjugated to a CPT.
