Abstract

Cell penetrating thyclotides (CPTs) significantly enhance the cellular uptake and endosomal escape of bioactive peptides.

  • CPTs are synthetic molecules designed to facilitate the entry of peptides into cells.
  • Peptides, despite their medicinal potential, often struggle to cross cell membranes effectively.
  • CPTs combine chiral tetrahydrofurans and polar sidechains, allowing for optimization in peptide delivery.
  • Two anticancer peptides with low bioavailabilities showed markedly improved cellular uptake when conjugated to a CPT.
  • Utilizing CPTs for cellular delivery may represent a novel approach for enhancing the effectiveness of bioactive molecules.
