Key Aging-Related Genes in Rheumatoid Arthritis Found by Computer Analysis and Confirmed in Mouse Immune Cells and Patients

Abstract

The derived nomogram showed robust diagnostic performance with an AUC of 0.988.

  • Three senescence-related hub genes (TNFAIP6, SLC2A3, RIPK2) were identified as key regulators in rheumatoid arthritis.
  • Hub genes exhibited strong correlations with myeloid cells, particularly macrophages.
  • Two distinct immunological subtypes of rheumatoid arthritis were identified.
  • RIPK2 was found to be upregulated in clinical samples, and its inhibition reduced inflammation and senescence-like changes in macrophages.
  • Preliminary data suggest that RIPK2 may influence these processes through the NF-κB pathway.
