Essence

A human centenarian-linked COL25A1 variant may promote longevity through collagen cleavage and immune-related signaling when modeled in C. elegans.

Evidence

In C. elegans, the col-99 S106L -mutant extended lifespan, required p38 MAPK and YAP-1, altered furin-mediated cleavage in vitro, and lifespan extension was reproduced by the cleaved extracellular COL-99 domain.

Caveat

The longevity effect was shown in worms with supporting in vitro cleavage data, not as direct evidence that the same variant extends human lifespan.
