Levels of p62 protein in spinal fluid linked to tau buildup, brain changes, and thinking problems in adults without dementia

Updated

Abstract

Elevated CSF p62/SQSTM1 levels are associated with poorer cognition and an increased risk of dementia (HR=1.78).

  • Higher levels of CSF p62/SQSTM1 correlate with increased tau pathology and elevated levels of sTREM2 and PGRN.
  • Decreased whole-brain and fusiform gyrus volumes are linked to elevated CSF p62/SQSTM1 levels.
  • Age modifies the relationship between p62/SQSTM1 levels, cognition, and whole-brain volume.
  • Significant associations between p62/SQSTM1 and cognitive function are primarily observed in middle-aged individuals.
  • Longitudinal analyses indicate a higher risk of developing dementia for participants in the highest quartile of p62/SQSTM1.
