  • Inflammatory signals from the gut and misfolded α-synuclein may activate the brain's immune response.
  • Activation of the cGAS-STING pathway in the brain could lead to chronic neuroinflammation.
  • Chronic inflammation may impair the brain's ability to clear waste, such as mitochondrial DNA and α-synuclein aggregates.
  • This impairment could create a cycle that enhances cGAS-STING signaling and sustains the disease process.
  • The triad model provides insights into the disease's progression, pathology spread, and clinical variability.
