The cGAS-STING pathway’s role in memory loss linked to Type 2 diabetes: from sensing damaged cell DNA to brain cell damage

Updated

Abstract

Type 2 diabetes mellitus is associated with elevated dementia risk through mechanisms beyond chronic hyperglycemia.

  • Mitochondrial dysfunction may initiate oxidative stress and impair the removal of damaged mitochondria.
  • The cGAS-STING pathway could detect abnormal mitochondrial DNA in the cytosol, leading to neuroinflammation.
  • Activation of the cGAS-STING pathway is linked to increased production of pro-inflammatory molecules that may disrupt brain function.
  • This pathway may compromise the integrity of the blood-brain barrier and hinder the formation of new neurons in the hippocampus.
  • Targeting the cGAS-STING pathway could represent a potential therapeutic strategy for cognitive decline associated with diabetes.
