Specially designed lipids control RNA assembly in mineral-rich environments

Updated

Abstract

A lead formulation of bisphosphonate lipid nanoparticle (BP-LNP) maximizes hydroxyapatite affinity while maintaining transfection potency.

  • Localized delivery of RNA therapeutics is often hindered by poor retention and rapid clearance at target sites.
  • The developed solid-phase transfection platform turns mineralized surfaces into bioactive reservoirs for nucleic acids.
  • BP-LNPs adsorbed onto mineral substrates resist fluid removal, enabling sustained RNA delivery to nearby cells.
  • This platform, termed MiTEX, shows versatility in delivering Cre recombinase mRNA and STAT3 siRNA for therapeutic purposes.
  • The approach may allow for transforming inert calcium phosphate materials into effective RNA delivery systems.
