Improving a hybrid RNA system to create circular RNA treatments

Abstract

A universal chimeric permuted intron-exon system has been developed to enhance RNA circularization efficiency.

  • The new system may improve the production of highly purified circular RNAs, which are promising for therapeutic applications.
  • CircRNAs were found to express low immunogenicity while maintaining potent protein translation capabilities.
  • Vaccines encapsulating circRNAs targeting human papillomavirus and SARS-CoV-2 demonstrated robust immune responses in mouse models.
  • This approach could facilitate the development of effective circRNA-based vaccines against tumors and viral infections.
