Cholesterol 25-hydroxylase (CH25H) linked to fat tissue inflammation in obesity and diabetes

Abstract

Increased levels of Cholesterol 25-Hydroxycholesterase (CH25H) are associated with metabolic inflammation in obese individuals.

  • Gene pathways related to inflammation were found to be elevated in obese diabetic individuals compared to non-diabetic subjects.
  • CH25H was significantly enriched in the stromal vascular fraction of human adipose tissue and highly expressed in specific immune cells.
  • A correlation was observed between Body Mass Index (BMI) and the levels of 25-Hydroxycholesterol (25-HC) in human visceral adipose tissue.
  • In mouse models, a high-fat diet increased Ch25h expression and 25-HC levels in adipose tissue.
  • Ch25h knockout mice showed reduced plasma insulin levels and improved insulin sensitivity despite becoming obese, linked to decreased immune cell infiltration in adipose tissue.
  • 25-HC was necessary for the expression of inflammatory genes in various adipose tissue cells.
