Abstract
A histone H3 lysine 37 to alanine (H3K37A) mutation promotes histone degradation and hyperactivates TORC1, leading to cytotoxicity upon TORC1 inhibition.
- Histone H3K37A mutation causes specific cytotoxicity when TORC1 is inhibited, unlike wild-type histone H3.
- H3K37A mutation results in enhanced histone degradation and exacerbated TORC1 hyperactivity.
- Transcriptome analysis reveals that H3K37A disrupts mitochondrial retrograde signaling, worsening mitochondrial dysfunction under TORC1 inhibition.
- Defective mitochondrial import of an oxidative phosphorylation subunit is linked to the observed cytotoxicity.
- Inactivation of reactive oxygen species can prevent the toxicity associated with H3K37A mutations.
