Ciliatoside A may help treat pneumonia caused by Klebsiella pneumoniae by improving cell cleanup through the SIRT1/PINK1/Parkin pathway

Abstract

Ciliatoside A (CA) significantly improved cell viability and reduced apoptosis in A549 cells infected with Klebsiella pneumoniae.

  • CA treatment decreased the expression of drug resistance genes in both Klebsiella pneumoniae and A549 cells.
  • Mitochondrial reactive oxygen species (mtROS) accumulation was reduced, while mitochondrial membrane potential and ATP production were enhanced by CA.
  • CA promoted mitochondrial autophagy and inhibited cell death associated with the NLRP3 inflammasome.
  • In a mouse model, CA alleviated pulmonary edema and tissue damage resulting from Klebsiella pneumoniae infection.
  • The SIRT1/PINK1/Parkin pathway was activated by CA, which was previously blocked following infection, suggesting a potential mechanism of action.
