Intestinal MRP2 expression shows a diurnal oscillation in mice, peaking at ZT6.

  • Bmal1 ablation reduces Mrp2 mRNA and protein levels, as well as transport activity.
  • Loss of Bmal1 disrupts the circadian rhythm of MTX pharmacokinetics and toxicity.
  • Dbp activates and E4bp4 represses Mrp2 transcription by binding to the same D-box element.
  • Bmal1 directly activates the transcription of Dbp and Rev-erbα, while negatively regulating E4bp4.
  • Positive regulation of Mrp2 by Rev-erbα is achieved through modulation of E4bp4.
